Butyl methanesulfonate
Butyl methanesulfonate (CAS 1912-32-9), also called methanesulfonic acid butyl ester, n-butyl methanesulphonate or NSC 36060, is a colourless to pale yellow liquid with a density near 1.107 grams per cubic centimetre at twenty degrees Celsius and a boiling point of about 81 to 83 degrees Celsius at four torr. It is the butyl member of the alkyl mesylate family, in which the sulfonate group is a very good leaving group and the butyl chain is transferred readily to nucleophiles including amines, thiols, phenoxides and carboxylates. The material is moisture sensitive, harmful if swallowed and it forms a mesylate ester with alkylating and mutagenic potential. It is therefore also supplied as a European Pharmacopoeia reference standard for controlling these genotoxic impurities in drug substances.
1. Alkylating reagent: used to introduce the n-butyl group into phenols, amines, thiols and nitrogen heterocycles in fine chemical, agrochemical and pharmaceutical synthesis as a milder and more selective alternative to butyl halides and dialkyl sulfates.
2. Genotoxic impurity reference standard: supplied as a pharmacopoeial reference substance for the detection, identification and quantification of alkyl methanesulfonate impurities in active pharmaceutical ingredients and intermediates.
3. Process chemistry: employed to study mesylate formation, purge factors and clearance during drug manufacturing, supporting toxicological risk assessment, control strategies and specification setting for potentially genotoxic impurities under regulatory guidance.
4. Handling and storage: harmful if swallowed, irritating to skin and eyes and potentially mutagenic; store cool and dry in a closed container away from water, bases and amines, use protective equipment and containment, avoid inhalation of vapour and treat residues as hazardous waste.
