Carbamimidic acid, methyl ester, sulfate
O-Methylisourea sulfate (CAS 29427-58-5), named in the CAS registry as carbamimidic acid methyl ester sulfate and also listed as O-methylisourea hydrogen sulfate or bisulfate, is the sulfuric acid salt of the reactive, volatile and strongly basic free base O-methylisourea. It is a white crystalline, water-soluble solid with the EINECS number 249-622-6, stable when kept dry and cool, and supplied at 99 percent purity for synthetic use. The molecule is the O-methyl imidate of urea and a powerful guanylating reagent: it transfers the amidine group to nucleophilic nitrogen atoms, converting amines, amino acids and aminoglycosides into guanidines and isoureas. Because the free base is difficult and hazardous to handle, the sulfate salt is the preferred commercial form for laboratory and industrial use.
1. Guanylation reagent: converts primary and secondary amines, amino acids and aminoglycosides into guanidines and isoureas, an essential transformation in pharmaceutical, peptide, carbohydrate and biochemical synthesis.
2. Pharmaceutical and agrochemical synthesis: used in the manufacture of guanidine-containing drugs, pesticides and herbicides, and in preparing creatine, creatinine, methylguanidine, agmatine and related nitrogen-rich compounds.
3. Heterocycle and intermediate chemistry: reacts with diamines, amino alcohols and aminothiols to give cyclic amidines, imidazolinones and thiazolines used as building blocks, protecting groups and organocatalysts.
4. Handling and storage: harmful by ingestion and inhalation and irritating to skin and eyes; keep the container sealed, dry and cool away from acids and oxidising agents, use protective equipment and ventilation, and avoid exposure to moisture, which liberates the volatile free base.
