Ethyl 2-acetate
Ethyl 2-(2-chloroethoxy)acetate (CAS 17229-14-0), also listed as ethyl 2-chloroethoxyl acetic acid or ethyl 2-chloroethoxy acetate, is a colourless liquid bifunctional ester containing a reactive primary alkyl chloride and an ethyl ester on a short glycol-like backbone. It is stored sealed, dry and refrigerated at two to eight degrees Celsius because the chloroether linkage is sensitive to hydrolysis and to prolonged heating, and it is supplied at 98 percent purity, soluble in common organic solvents including dichloromethane and dimethylformamide. The molecule offers two orthogonal reactive handles: the chloride undergoes nucleophilic substitution with amines, thiols, alkoxides and carboxylates, while the ester can be hydrolysed, reduced, transesterified or converted to an amide. This makes it a compact and flexible building block for medicinal and process chemistry.
1. Pharmaceutical intermediate: used to construct ethoxy-linked and morpholine-containing drug fragments, including intermediates for analgesics, antihistamines, cardiovascular agents, antitussives and central nervous system therapeutics.
2. Linker and spacer chemistry: the chloroethoxy ester provides a short, flexible, hydrophilic linker for conjugating drugs, fluorescent probes, peptides and polymers in bioconjugation, prodrug and delivery-system design.
3. Synthetic transformations: the alkyl chloride can be displaced to give amino, thio, azido or alkoxy ethers, while ester manipulation yields carboxylic acids, amides, hydrazides and alcohols for further elaboration into heterocycles.
4. Handling and storage: irritating, moisture sensitive and harmful if swallowed; keep sealed, dry and refrigerated between two and eight degrees Celsius, avoid contact with strong bases that may promote elimination of hydrogen chloride, and use standard protective equipment with adequate ventilation.
