Kanamycin A sulfate
Kanamycin A sulfate (CAS 25389-94-0), also catalogued as kanamycin sulfate or kanamycin A monosulfate, is the sulfate salt of kanamycin A, an aminoglycoside antibiotic originally isolated from the soil bacterium Streptomyces kanamyceticus. It is a white to off-white, highly water-soluble powder that is stable in the dry state at 98 percent purity or higher and is active against a broad range of gram-negative and gram-positive bacteria. The molecule consists of two amino sugars glycosidically linked to a 2-deoxystreptamine core, giving it a strongly cationic character at physiological pH that drives binding to the bacterial 30S ribosomal subunit and thereby inhibits protein synthesis and leads to bacterial cell death. It is a mainstay of both clinical antimicrobial therapy and laboratory microbiology.
1. Antibacterial therapy: an injectable and oral aminoglycoside used to treat serious gram-negative infections, as a second-line antituberculosis agent, and in veterinary medicine and topical or ophthalmic preparations.
2. Molecular biology selection marker: the kanamycin resistance gene is one of the most widely used selectable markers for plasmid transformation, cloning, recombinant protein expression and genome engineering in bacteria, yeast and mammalian cells.
3. Cell culture and microbiology: added to culture media to suppress bacterial contamination and to maintain selection pressure in recombinant strain construction and protein production systems.
4. Handling and storage: store the dry powder tightly closed and protected from light and moisture at two to eight degrees Celsius; aqueous solutions remain stable after sterile filtration, and the material is a potential sensitiser that should not be inhaled.
