Triphenyltetrazolium Chloride
Triphenyltetrazolium chloride is a water-soluble tetrazolium salt, supplied as a white to pale yellow crystalline powder at 98% purity.
In living tissues the colourless salt is reduced by dehydrogenase activity to formazan, an intensely red, water-insoluble pigment, so the intensity of the colour reports directly on metabolic activity and cell viability. This simple, robust chemistry has made it a standard reagent for viability assays, seed testing and histochemical staining.
The solid is light-sensitive and should be stored dry, cool and protected from light; aqueous solutions are best prepared fresh or kept refrigerated in the dark.
• Cell viability assays: reduced by metabolically active cells to red formazan, allowing colorimetric quantification of proliferation and cytotoxicity.
• Seed and tissue viability testing: the classic tetrazolium test for germination capacity in seeds and for viability of plant and animal tissue slices.
• Histochemistry and microbiology: used to localise dehydrogenase activity in tissue sections and to detect microbial growth and biofilm activity.
• Analytical indicator: applied in redox studies, in the detection of reducing compounds and as a substrate in enzyme-activity measurements.
